DNA integrity at the heart of product safety

The integrity of the genetic information carrier is essential to human health. DNA impairment can lead to diseases such as (lung) cancer, vascular disease and premature aging.  Every substance or ingredient intended for human consumption is therefore subjected to ever-greater control. The integrity of the DNA is of particular concern and thus a genotoxic risk assessment is often required or may even be mandatory.

Risk assessment procedures hitherto heavily rely on the use of experimental animals including the pharmaceutical, cosmetics and chemical industry, regulatory and basic sciences. In 2011, a report by the European Commission on the statistics of the number of animals used for scientific purposes by European countries showed that nearly 11.5 million animals were used for scientific and other experimental purposes in the European Union. Around 80% were rodents and rabbits; 61% of the total number of animals were mice. The importance of animal research in certain areas is uncontested, however, there exits convincing evidence for the need of a global and significant reduction in use of animal experimentation for quality control purposes.

Growing awareness for alternative quality control testing

The European Society for Alternatives to Animal Testing (EUSAAT) actively promotes the protection of experimental animals according to the 3Rs-Principle (refine - reduce - replace) developed by Bill Russel und Rex Burch in 1959. EUSAAT is serving as European 3Rs platform by offering expertise and networking, and supporting educational programs in close cooperation with NGOs, regulators, research institutions and industry to improve and implement humane experimental techniques as wells as advanced in silico und in vitro methods (www.eusaat-congress.eu). 3T’s latest product, AUREA gTOXXs, an automated (3D) cell based diagnostic platform identifies candidates with DNA damaging effects directly on human cells, cell culture or 3D tissue culture. Animal-free DNA-based risk assessment by AUREA gTOXXs in HepaRGTM model cells

The HepaRGTM cell line from Biopredic International, France, is an excellent model for mapping the metabolic response of human hepatocytes to chemicals and provides a valuable alternative system to study liver toxicities (www.biopredic.com). Bioactivation through drug or agent metabolism in liver cells is frequently suspected as an initiating event in many (geno)toxicities as is the case for the extremely harmful mycotoxin, aflatoxin B1 from Aspergillus flavus. Recently, the applicability of the HepaRGTM cell line for gTOXXs based genotoxicity studies of drugs with the need for bio activation by liver cells has been demonstrated by showing the harmful effect of aflatoxin B1 on the DNA of HepaRGTM cells. The additional information on DNA repair activity by AUREA gTOXXs provides unique data on the validity and regeneration of the damaged cells.

Diesel emission tests on monkeys or even humans, as well as animal experiments for marketing of new food, cosmetic, or medical ingredients are becoming increasingly superfluous. Whether it’s sunscreen, nanoparticles, superfoods or chemotherapeutics - AUREA gTOXXs supports the release of safe products to the market without relying heavily on animal models.
